Abstract

Enhanced Selectivity for Absolute Protein Quantification by LC-MS3, presented at the 59th ASMS conference on Mass Spectrometry and Allied Topics, June 5-9, 2011 in Denver, CO.

Objective: Evaluate the use of fast MS3 scans for protein quantification in terms of assay LOQ, precision and accuracy.

Approach:

  • compare the performance of SRM and fast MS3 scans in the quantification of UGT isozymes in liver microsomes
  • Quantify UGT isozymes using unique surrogate tryptic peptides

Conclusion:

  • The performance of fast MS3 scans is comparable to SRM scans in terms of precision and accuracy
  • The use of fast MS3 scans allows more MS3 transitions to be monitored in one acquisition period compared to the default MS3
  • Absolute quantification of human UGT enzymes using Fast MS3 scanning can provide a significant enhancement in assay selectivity
